Inclusion criteria

The key inclusion criteria are -

  • Diagnosed HR-positive (ER >1%) HER2-negative locally advanced or metastatic breast cancer
  • ≥1 lesion (measurable and/or non-measurable) accurately assessable at baseline and suitable for repeated assessment as per RECIST v1.1
  • Germline or tumor loss of function mutation in BRCA1, BRCA2, or PALB2 based on CLIA/CAP-certified/locally accredited laboratory results or the central tumor test, respectively
  • FFPE tumor sample for genetic tissue testing, additional biomarker analysis, and companion diagnostic development

Exclusion criteria

The key exclusion criteria are -

  • Prior systemic anti-cancer therapy (excluding ET) up to 28 days before randomization
  • Disease progression ≤84 days after last neoadjuvant/adjuvant chemotherapy
  • Disease progression ≤365 days from last dose of:
    • PARPi and/or platinum agent for eBC
    • CDK4/6i in the adjuvant setting
    • Oral SERD including camizestrant
  • Any clinically important abnormalities in resting ECG and/or resting heart rate <55 bpm for participants either taking heart rate reducing concomitant medications or with a history of heart diseasea

aStroke, or uncontrolled coronary heart disease, or dysrhythmia.

Target Overview

Poly [ADP-ribose] polymerases (PARPs) are a group of proteins known to be involved in the repair of single-strand breaks in DNA.1

A healthy cell’s DNA undergoes damage every day, with approximately 10,000 spontaneous single-strand breaks that require repair.1

Inhibition of PARP1 with AZD5305 leads to the trapping of PARP bound to DNA single-strand breaks, stalling of replication forks, their collapse and the generation of DNA double-strand breaks. In tumor cells that already have compromised DNA repair mechanisms (such as loss of BRCA1 or BRCA2 function, affecting double-strand break repair), inhibition of PARP leads to excessive accumulation of DNA damage and may cause death of the cell.1,2

Compound Overview

Saruparib (AZD5305) is an investigational, new generation PARP inhibitor. It is designed to selectively inhibit and trap PARP1, enabling efficacy, while broadening the opportunity for clinical benefit by allowing combination approaches with different treatment options.3,4
